Objectives:

Applying the CRISPR/Cas9 coupled recombineering strategy to assemble tracts of multi-kilobase human synthetic sequences in the E. coli chromosome, subsequent assembled DNA recovery from the chromosome and genomic replacement in mouse ES cells.
